Comparação entre técnicas de coloração de raízes de soja infectadas por Heterodera glycines, Meloidogyne incognita e Pratylenchus brachyurus

Resumo

Experiment was conducted in a greenhouse with the purpose to evaluate the effectiveness of two root staining techniques on soybean cultivar Emgopa 313, infected by the three main nematodes of the crop: Heterodera glycines, Meloidogyne incognita and Pratylenchus brachyurus. Three evaluations were performed at 10, 17 and 25 days after planting in naturally infested soil. The technique using bleaching with NaOCl and acid fuccin and the technique that employs lacto-glycerol and acid fuccin were both effective in staining nematodes, allowing its observation in the root tissue. Because of longer duration of the life cycle of P. brachyurus it was not possible to view these nematodes in root the tissue at the first evaluation using the technique of bleaching and staining with acid fuccin, and at the second evaluation using both techniques. For young tissues the technique that employs bleaching is more indicated to facilitate the nematode observation, since it uses less stain which helps the process of discoloration.
