Morphological and histological characteristics, phytochemical tests, ash and reducing sugars contents are descripted. The results showed steroids/ triterpenes, sugars reducers, starch, mucilage and saponinas presence; the tenor of total ashes was of 2,21%, and insoluble in acid of 0,63%; the humidity of 6,42% and the tenor of sugars reducers was of 2,77%.Item Análise farmacognóstica do pó e avaliação do efeito do extrato etanólico das flores da calendula officinalis l. cultivadas no Brasil no sistema imunológico(Marize Campos Valadares, 2004-12) Parente, Leila Maria Leal; Matos, l. (Asteraceae) is an annual herbaceous from Mediterranean area and it was adapted in Brazil. Lots of factors related to cultivation, can affect in a significant way the quality and the quantity of the active principles produced by the plants. In this work, pharmacognostic evaluation of the flowers of C. officinalis from Brazil (Paraná), was accomplish for the quality control of the botanical material. The plant, well known a long time ago, has anti-inflammatory and healing activities and several other effects. It is present in many commercialized therapeutic formulations. In this work, an ethanolic extract of the flowers of the cultivated C. officinalis (EEC) in Brazil was gotten and was become an evaluation of the effect of the EEC in the immunologic system, verifying if the extract also gotten it would present the same immunomodulatory activity for isolated fractions of the plant cultivated in the Europe and Asia.Item O uso de cultura clonal de precursores ematopoiéticos na investigação do potencial mielossupressivo de novas substâncias(Marize Campos Valadares, 2004-12) Valadares, Marize CamposThis article is a short review of the employment of the hematopoietic progenitor clonogenic assay, in particular the clonogenic assay for hematopoietic progenitors of granulocyte/macrophage (“Colony Forming Unit - Granulocyte Macrophage”, CFU-GM) in the investigation of myelotoxicity.Item Determinação de ginkgoflavonóides por cromatografia líquida de alta eficiência em matériasprimas e produtos acabados(Marize Campos Valadares, 2004-12) Bara, Maria Teresa Freitas; Cirilo, Hérica Núbia Cardoso; Oliveira, Valéria dePharmaceutical preparations containing Ginkgo biloba L. have large market. Pharmacological and clinical survey on EGb761 extract have demonstrated application on peripheral circulatory and cerebrovascular insufficiency. The Ginkgo extracts contain as active compounds flavonoids and terpene lactones. In this work the quality of the standardization extracts and of the preparations containing Ginkgo was evaluated. The method for quantification of Ginkgo flavonoids was liquid chromatography and data showed to be simple, rapid and adequate to this analysis. The analytical results demonstrated that 33% and 92%, respectively, plant extract and capsule were not in agreement with the specifications. This analysis showed valuable for herbal medicines and phytomedicines quality control.Item Validação analítica em CLAE-UV para monitoração plasmática da metilprednisolona e do succinato de metilprednisolona(Marize Campos Valadares, 2004-12) Cunha, Luiz Carlos da; Ramalho, Pedro Ivo Sebba; Rezende, Kênnia Rocha; Marinho, Carlos H. C.; Oliveira, Emílio Costa de; Silva, Nílzio Antonio daThe HPLC-UV analytical method validation aimed the quantification of methylprednisolone (MP) and its succinate sodic ester (MPSS) in human plasma of patients with lupus. Triancinolone (500 g/mL) was used as internal standard the standards MP and MPSS were spiked in plasma in the range of 10 and 1.0 g/mL. The chromatographic conditions employed an isocratic mobile phase (acetonitrile:water:acetic acid 47:50:03), flow rate of 0.8 mL/min and a RP18 column (25 cm and 0.4 mm i.d.) coupled to an UV detector set at 254 nm. The parameters of analytical validation are in compliance with international regulations and with the Brazilian sanitary legislation. Beyond excellent linearity (RMP = 0.99933; RMPSS = 0.99638), it showed high intraday and interday precision (< 7%), good accuracy (94,0 ± 2.3%), adequate limit of quantification (0.25 g/mL) and good recovery (> 85%). Additionally, the technique have shown to be robust, steady, selective and can be used in the dosage of methylprednisolone in patients under pulsotherapy and also in situations of acute poisoning and therapeutical monitoring.Item Emprego da técnica de HRGC na identificação da isotretinoína e seus produtos de degradação em medicamentos(Marize Campos Valadares, 2004-12) Diniz, Danielle Guimarães Almeida; Alves, Carina Pimentel Itapema; Lima, Eliana Martins; Antoniosi Filho, Nelson RobertoIsotretinoin or 13-cis-retinoic acid is a member of the large group of Vitamin A related compounds. Due to its effect on regulating cell differentiation it has been used for the treatment of cystic and nodular acne and also as an inhibitor of neoplasic cells proliferation. This paper describes a gas chromatography method used for the assay of isotretinoin in its isolated form and in pharmaceutical formulations. Results showed that the method exhibited a high efficiency to determine the quality of isotretinoin in raw materials and in pharmaceuticals.Item Validação de técnica analítica em cromatografia em camada delgada comparativa para identificação de fármacos anorexígenos sintéticos em produtos fitoterápicos(Marize Campos Valadares, 2004-12) Azeredo, Flaubertt Santana de; Guimarães, Renato Inácio; Paula, José Realino de; Cunha, Luiz Carlos daThere are reports of synthetic substances not mentioned in the composition of slimming phytotherapics freely marketed being they responsible for the apparently miraculous effects of these formulations. These substances act in the Central Nervous System (CNS) with sale and use restrictions. Due the need to identify those adulterants, a comparative thin layer chromatography technique (TLC) was validated for drugs (1mg/mL in methanol solution) used in the treatment of obesity (femproporex, mazindol, diethylpropion, fenfluramine, hydrochlorotiazide, furosemide, fluoxetine, benzocaine, diazepam, midazolam, flurazepam, bromazepam, nitrazepam, clordiazepoxide, caffeine, phenolphthalein). Analytical interference with 20 mg of standard phytodrugs (Cynara scolynmus, Aloe vera, Peumus boldus, Baccaharis sp, Rhamnus purshiana, Centella asiatica, Garcinia sp, Chorella pyrenoidosa, Maytenus ilicifolia, Spirulina maxima, Fucus sp., Paullinia cupana, Pfaffia paniculata, Amorphophallus konjac, Solanum paniculatum, Passiflora alata Cassia angustifolia) extracted with 5 mL of methanol was tested. Analytical technique of TLC was developed in silica gel G60’s plate, 0.5 mm, 20x20 cm, with mobile phase and sequence of detection reagents for substances of acid/neutral and basic characters. The Rf’s values and developed spot colors were considered. The procedure for the "natural products” was performed, whose results were compared to the patterns, with repetitions for the suspicious products. It was concluded that the technique is selective, fast and simple and have low cost. It could be developed at any analytic laboratory for the identification of the synthetic anorexic drugs in phytomedicine composition.Item Farmacogenoma do fluorouracil: análise de polimorfismo no gene da diidropirimidina desidrogenase(2005) Provin, Mércia Pandolfo; Araújo, Rogério Agenor; Nascimento, Lorraine Lemes; Goulart Filho, Luiz RicardoItem Calcium carbonate reduces iron absorption from iron sulfate, but not when iron is presented as an organic complex(2005-03) Conceição, Edemilson Cardoso da; Machado, Alexandre Alves; Izumi, Clarice; Freitas, Osvaldo deExperimental and epidemiological evidences have demonstrated that calcium inhibits iron absorption; calcium carbonate being one of the most effective calcium sources to reduce iron absorption from dietary origin or from iron sulfate. In the present work, the short-term effect of calcium from calcium carbonate on iron absorption was studied in rats, using different iron compounds (monosodium ferric EDTA, iron-bysglicine, iron peptide complex) with iron sulfate as a control. Eighty (80) animals were divided into groups of 10 animals each with homogeneous weight. After 18h fast, the animals received by gavage 5 mL of a dispersion containing one of the iron compounds (1mg Fe/kg body weight), concomitantly or not with calcium carbonate at a molar ratio of 150:1 (Ca/Fe). Two hours after the administration, the animals were sacrificed and blood was collected for serum iron determination (iron transfer rate from intestinal lumen to blood compartment). Additionally, the intestines were collected for soluble iron determination (available iron). The results demonstrated that calcium ion from calcium carbonate inhibits the iron absorption from iron sulfate, but not from organic iron (di- or trivalent) complexes.Item Tentativa de suicídio com o uso de medicamentos registrados pelo CIT-GO nos anos de 2003 e 2004(Marize Campos Valadares, 2005-06) Rios, Diogo Passos; Bastos, Fernando M.; Cunha, Luiz Carlos da; Valadares, Marize CamposAccidental ou proposital administratrion of potentially toxic agents is related to severe intoxications. Considering all classes of intoxications (minor to severe) the majority are caused by drugs, like as antidepressant, analgesic, anti-anxiety and anticonvulsant agents. The suicide attempt is very common, instead of clinical or forensic misdiagnostic of such psychiatric disturbs. In this work we evaluated and compared suicide attempt, caused by medicine ingestion, recorded on Center of Toxicological Information of the Goias State (CITGO) over the years 2003 and 2004. In such period, 597 cases of poisoning were related to medicines. In the period of 2003, 237 cases had been notified, while that in the year of 2004 had been notified 360 cases, 1.5 times more than the previous year. The psychiatric drugs, analgesics and non-steroidal antinflamatory drugs (NSAIDS) had been responsible for more than 60% of all the cases of suicide attempts. About 80% of the medecine suicide attempt cases it occurred in the feminine sex, mainly in the age range of 21 the 30 years, with low index of lethality (0,84 - 2.49%).Item Annona coriacea induz efeito genotóxico em camundongos(Marize Campos Valadares, 2005-06) Fagundes, Flávio A.; Oliveira, Leonardo B. de; Cunha, Luiz Carlos; Valadares, Marize CamposThis study was designed to evaluate the genetic toxicity of the Annona coriacea using the bone marrow erythrocyte micronucleus test. The results demonstrated that the seed alcoholic extract of this plant, in different doses used (50, 100, 200 or 400mg/kg) induced a significant increase in micronucleus frequencies in bone marrow erythrocytes of mice, when compared to the negative control. In addition, this increase was similar to those found in the group of mice exposed to the positive control (cyclophosphamide 250mg/kg). These results suggest that this plant, in this experimental condition, has a significant potential for induce DNA damage.Item Biotransformaçâo do 1,8–cineol por bactérias livres e imobilizadas(Marize Campos Valadares, 2005-06) Almeida, Ludimila P.; Ferri, Pedro Henrique; Paula, José Realino de; Santiago, Mariângela FontesThe essential oil of leaves of the Eucalyptus microcorys F. Muller collected in Silvânia-GO, possesses 87% of 1,8-cineole. This monoterpene is one of the main composed of the essential oils of Eucalyptus globulus Labill and they are of great interest for the industries, mainly the perfumeries, food and pharmaceutical. The objective of this work was the hydroxylation of 1,8-cineole for free and immobilized bacterias. The hidroxilation of the molecule of 1,8- cineole is important for obtaining new aromas. The essential oil of Eucalyptus microcorys also presented inhibition on the growth of Staphylococcus aureus ATCC 6538 and the hydroxylation of the molecule of 1,8-cineole can increase this activity antibacterial. In this work it was tested the Pseudomonas putida, Azotobacter vinelandii and Pseudomonas fluorescence bacterias. However the Pseudomonas putida went to that presented a significant result and facilitated the biotransformation of the composition 1,8-cineole. The main culture used for biotransformation (MMB) was: 0,5g of glucose, 0,2g of (NH4)2SO4, 0,4g of KH2PO4, 0,6g of Na2HPO4, 0,2g of MgSO4, amount lines of Fe, B, Mn, Zn, Cu and Mo in 100 mL of distilled water. After 60-65 hours to 30oC, pH 6,5 and 180 rpm 100 μL the 1,8-cineole was added in main MMB and was maintained at the same temperature conditions, pH and agitation, for periods of 48-120 hours. The solvent that showed more efficient for extraction of 1,8-cineole was the ethyl acetate. The ideal revelator for the composition 1,8-cineole at thin layer chromatography was a solution of 10 mL of H2SO4 concentrated, 45 mL ethanol, 1g of vanillin and 10 mL of distilled water. The mobile phase used at the chromatography was 1:3 of ethyl/hexano acetate. The results of biotransformation were investigated through the technique of GC/MS. The bacteria Pseudomonas putida got transform the substratum 1,8-cineole in two new composed: exo-2-hydroxycineole and the 1- hydroxy-p-menth-3-one.Item Avaliação clínica toxicólogica preliminar do viticromin® em pacientes com vitiligo(Marize Campos Valadares, 2005-06) Leão, Anatel Rocha; Cunha, Luiz Carlos da; Parente, Leila Maria Leal; Castro, Lia Cândida Miranda de; Chaul, Aiçar; Carvalho, Helena Eterna de; Rodrigues, Vânia B.; Bastos, Marciley AlvesThe Brosimum gaudichaudii Trécul (Moraceae) is a very common herb in brazilian vegetation called “Cerrado”. This plant is used in the ethnopharmacology for the treatment of the vitiligo. In this plant are founded the furancoumarins, mainly psoralen and bergapten which have a photosensibility property responsible for the dermal repigmentation effect. The Brosimum gaudichaudii is a main active component of a medicine called as Viticromin®, which is made from the inferior part of the stem and the roots of the plant. Considering the low toxicity of the Brosimum gaudichaudii’s roots extract in the pre-clinical toxicological studies, in this study we investigated same parameters of the clinical toxicology of the Viticromin® in vitiligo’s patients on therapy with this drug. The results showed that this drug produced no significant alteration on the aspects investigated in patients under the drug regimen studied.Item Estudo da ligação cruzada induzida pelo formaldeído em cápsulas de gelatina dura(Marize Campos Valadares, 2005-12) Zampieri, Ana Lúcia Teixeira de Carvalho; Lima, Eliana Martins; Diniz, Danielle Guimarães AlmeidaFormaldehyde induced cross-linking in gelatin can be used to promote gastric-resistance in gelatin based oral drug delivery systems. However, thorough evaluations for each drug class must be performed before establishing its use for this purpose. In this work, formaldehyde induced cross-linking in hard gelatin capsules was investigated. Capsules were immersed in hydroalcoholic (70-90%) solutions containing several different concentrations of formaldehyde. Cross-linking was evaluated by the determination of free amino groups in the samples by UV spectrophotometry. Residual formaldehyde was determined by HPLC. Cross-linking reaction was strongly influenced by water and alcohol concentrations in the solutions used for the treatment of the capsules. Residual formaldehyde was within acceptable range. However, repeated analysis during a short term storage of the capsules proved that the cross-linking continues as long as any residual formaldehyde is present, leading to a much higher resistance than what would be convenient for the oral drug delivery.Item Análise microbiológica de matérias primas e formulações farmacêuticas magistrais(Marize Campos Valadares, 2005-12) Andrade, Flávia R. O.; Souza, Aline A.; Arantes, Maria do Carmo Batista; Paula, José Realino de; Bara, Maria Teresa FreitasIn agreement with Good Manufacturing Practice, pharmaceutical products must have a quality according to specifications set by official compendia assuring their use. The microbiological quality of raw material employed in pharmaceutical and cosmetic formulations is very important to achieve efficacy and safety. In this work it was done the heterotrophic bacteria count, the total viable aerobic count, yeasts and moulds count, detection of pathogenic bacteria in samples of purified water, plant raw materials, natural products and in formula base of creams, lotions and gels. The applied methods and the specifications were based on Pharmacopeas or World Health Organization.The results showed that 86.3% of analysed samples were according to the parameters. The microbiological quality of deionizated water demonstrated a problem to pharmacies and it was verified that 65% of them were above the specifications. It was found that 97.17% of the formulations were with microbial counts in the permited limit. The plant raw materials and natural products showed values of 3.51% and 10.5%, respectively in disagreement with the specifications. The obtained data demonstrated that the Good Manufacturing Practice applied to pharmacies contribute to microbiological quality of the pharmaceutical products, however, critical points must be solved, as the quality of the deionizated water used must be improved.Item Análise da publicidade de medicamentos veiculada em Goiás - Brasil(Marize Campos Valadares, 2005-12) Freitas, Johnathan Santana de; Zampiere, Ana Lúcia Teixeira de Carvalho; Aquino, Aline Teixeira de; Lobo, Lina Monteiro de Castro; Montes, Patrícia Regina; Soares, Arthur; Silva, Érica T. da; A. Neto, João da C.; Araújo, Mariana de Paiva; Tresvenzol, Leonice Manrique Faustino; Prudente, Luciana Resende; Lima, Dione MarçalMedicines are special products, whose advertisement inserts require different care relating to their ethic and moral aspects, once they have highly specific characteristics and their use is based on a precise diagnosis of a situation that causes risk to health. In this regarding, regulatory agencies established a specific legislation (RDC n° 102/00), set in legal parameters to medicines advertisements. The aim of this work was to evaluate the advertisements of medicines in Goias-Brazil based on the current legislation. The captation of irregular medicine advertisement inserts was performed between November/2004 and September/2005. The analysis followed the guideline “Roteiro de Verificação de Conformidades das Publicidades e Propagandas de Medicamentos”, present in the “Manual de Monitoramento de Propaganda de Produtos Sujeitos à Vigilância Sanitária” (MS, 2005). The results showed most of advertisements were related to over the counter medicines (OTC) (88,63%), followed by medicine prescription (8,33%) and controlled prescription (3,04%). The advertisement flyers were identified as the principal responsible for the propagation of irregular inserts (81,06%), followed by TV (14,40%) and radio (4,54%). In the 132 inserts analysed were detected many non-conformities with legislation and the absence of information about cares and warnings was observed in the majority of them. The present work demonstrated the importance of the medicines advertisements build based on the current legislation, assuring the defense of the population´s health.Item Análises preliminares de substâncias viscosas em espectrômetro de infravermelho utilizando suporte alternativo(Marize Campos Valadares, 2006) Soares, Lillian Amélia; Gil, Eric de Soza; Rezende, Kênnia RochaInfrared spectroscopy is a very used technique for structural elucidation of organic compounds. However, some accessories used on sample preparation are somehow expensive. In this work, it was investigated an alternative procedure for qualitative analysis of viscous substances using PVC film like support. Analysis of lanolin anhydrous and vitamin E were realized in KBr discs and PVC film comparatively to literature. The main advantages of this method rely on running samples spending less time, at low-cost support cell obtaining confident spectra.Item Avaliação de toxicidade aguda: estratégias após a “era do teste DL50“(Marize Campos Valadares, 2006) Valadares, Marize CamposThe present work discuss about the evaluation of the acute systemic toxicity post deletion of the LD50 test (median lethal dose). The three alternative animal tests now employed, the Fixed Dose Procedure, the Acute Toxic Class Method and the Up and Down Procedure, give rise to significant improvements in animal welfare. Although, the total replacement of animal tests it is not possible yet, basal cytotoxicity in vitro has been used as adjunct to these alternative animal tests to select initial doses, thus reducing the numbers of animals used during the evaluation of acute systemic toxicity.Item Estudo imunohistoquímico do cerebelo de ratos wistar submetidos à exposição aguda ao etanol no 12º dia de vida intrauterina(Marize Campos Valadares, 2006-06) Souza, Aline Gomes; Rodrigues, Humberto Gabriel; Vieira, Clarissa Maria Serpa; Mateus, Marcus Vinícius Chaves; Aversi-Ferreira, Tales AlexandreThe effects of the chronic use of ethanol during pregnancy on the morphology and function of the neural system are known in literature, but the effects of the acute use of ethanol at critical moments of pregnancy still need to be clarified. The chronic prenatal exposure to ethanol have been associated to several anomalies in fetus and new-born rats, as alterations in the neuronal density of the piriform cells of the cerebellum, as known as Purkinje cells. In this paper, we have studied the effects of the acute exposure to ethanol at the 12th day of pregnancy of Wistar female rats on the morphology of the cerebellum in the brood. There was no decrease in the granular precursors and Purkinje cells, as occurred in the fetal alcoholic syndrome produced by the chronic exposure of pregnant rats to ethanol, which could be explained by the fact that the ethanol exposure happened after borning of the Purkinje cells, which constituted an adequate media to migration and surviving of the granular cells. The ethanol’s acute effects in begin of the cerebral cortical construction, demonstrate high severity on this tissue, but no in this date, can’t be observed, in this work, the same deleterious effects seen on cerebral cortex in cerebelar cortex.
