The animals had been kept in extensive conditions, with feeding the grass, receiving mineral supplementation and serving a herd with 3,782 females of some categories (heifers and cows). Data enclosed measured of scrotal circumference and evaluation of the semen collected by eletroejaculation and evaluated the physical characteristics (volume, mass activity, motility, strong), morphologic (major, minor and total defects) and spermatic structure integrity (integrity of plasmatic membrane, acrosome and chromatin) in this period had been carried through that preceded three consecutive mating season (2003, 2004 and 2005). The data had been submitted analyze of variance, the averages compared for test SNK and the coefficient of correlation of Pearson was calculated using the SAS. The bulls presented scrotal circumference of 33.8 ± 2.8cm, 15.6 ± 8.5% of total spermatic defects, 40.1 ± 23.1% of gametes with normal membrane and acrosome and 8.8 ± 3.8% of fragmented chromatin. The relations between scrotal circumference, physical and morphologic characteristics, spermatic integrity and the reproductive performance of the herd had been low and not significant (P< 0.05). In sexually mature bulls the integrity of plasmatic membrane, acrosome and nuclear chromatin had not been the most frequent trouble of the spermatic morphology.Item Lesões de isquemia arteriovenosa e reperfusão em jejuno de equinos: imunoistoquímica de proteínas de junção e histopatologia(Universidade Federal de Goiás, 2007-12-31) NASCIMENTO, Aline Machado Rapello do; ARAÚJO, Eugênio Gonçalves de; http://lattes.cnpq.br/3919777570059928; BRITO, Luiz Augusto Batista; http://lattes.cnpq.br/3212353728321020To evaluate whether complete arteriovenous ischemia followed by reperfusion of mesenteric blood flow aggravates lesions involving small intestine of horses, β--catenin and occludin expressions were measured in jejune epithelia of seven equines, without definite breed. The animals had been submitted to aseptic ventral midline celiotomy and five jejune segments collected corresponding to a baseline segment, a five minutes segment subjected to ischemia, and three segments collected after being subjected to five, sixty and one hundred and twenty minutes of tecidual reperfusion, respectively. Experimentally induced lesions created by total arteriovenous occlusion followed by reperfusion were similar to naturally acquired lesions, as much in the period of ischemia as in the aggravation of the injuries observed during reperfusion. Changes that occur when ischemic intestine is reperfused are progressive, and include mucosal, submucosal, and serosal edema; polymorphonuclear and mononuclear cell infiltrates; and alterations in vascular patterns. Imunohistochemistry analyses have shown strong staining baseline segments for β-catenin in all animals and its progressive lack of staining during ischemia and reperfusion treatments. That indicates progressive lack of intercellular adherence. Similar results of specific decreases of total expression of Occludin, a tight junction protein, were observed. This protein is involved in regulating the movement of solutes in the paracellular pathway and also plays an important role in the maintenance of cell polarity. The reduction of expressions of both intercellular proteins measured directly demonstrates β-catenin injury due to progression of edema during ischemia and its aggravation during reperfusion and also shows an increase of the permeability of paracelular pathways and its related consequences to degradation of Ocludina.Item Produção lacrimal e densidade de células caliciformes conjuntivais em cães da raça SHIH-TZU(Universidade Federal de Goiás, 2008-02-28) LIMA, Aline Maria Vasconcelos; ARAÚJO, Eugênio Gonçalves de; http://lattes.cnpq.br/3919777570059928; BRITO, Luiz Augusto Batista; http://lattes.cnpq.br/3212353728321020; CHAVES, Nilo Sérgio Troncoso; http://lattes.cnpq.br/9218281815823682Considering the relevance of tear production and tear quality to the conservation of the corneal health, the present study aimed to evaluate tear production and conjunctival goblet cells density in Shih-tzu dogs. Thirty five grownup Shih-tzu dogs, including 33 females and 2 males, were subjected to Schirmer tear test 1 (STT-1) and Schirmer tear test 2 (STT-2). Among these dogs, fourteen females and one male were underwent to conjunctival biopsy for histologic evaluation of goblet cell density (GCD) and goblet cell index (GCI). The STT-1 mean values found were 19,66 ± 7,3 mm/min for left eye (LE) and 21,97 ± 5,7 mm/min for right eye (RE). The STT-2 mean values were 10,71 ± 6,1 mm/min and 9,14 ± 4,78 mm/min for LE and RE, respectively. The GCD mean values found were 13,64 ± 3,44 goblet cells/50 epithelial cells for LE and 13, 64 ± 4,07 goblet cells/50 epithelial cells for RE. The GCIs correspondent were 0,27 ± 0,069 and 0,27 ± 0,081 for LE and RE, respectively. There was significant difference between LE and RE STT-1 values. There was not significant difference between eyes for STT-2, GCD and GCI values. STT-1 and STT-2 values did not influence significantly GCI values. STT-1 and STT-2 values are within of the pattern considered normal, but the GCD and GCI mean values are lower than those values described in literature for healthy dogs of other breeds. The conjunctival specimens histological evaluation was accepted as convenient, which demonstrated squamous metaplasic areas in 83,3% of the specimens. They may be used in exploration of low-quality pastures, without great investments, when other breeds could show a low productivity or even survival difficulties. This genetic potential runs risk to extinction due to its replace by more productive breeds. The Curraleiro cattle resistence to illness is popularly known and many works has been published about it, although its immunologic system physiology is completely unknown. Such features are plausible reasons for the preservation of this potentially genetic resource. So, the mean aim of this study was to establish an immunological profile by marking and quantification of T and B lymphocytes in Curraleiro breed with immunocytochemistry. Thus, it was used 116 bovines, male and female, with different ages from two farms situated at Goiás State, Brazil. The animals were allotted in groups according to age, sex and origin. Blood samples were collected and processed in accordance with immunocytochemistry standard technique using lymphoid markers species-specific, anti-CD3 (MM1A BoCD3) and anti-LB (LCTB16A clone B-B14), for T and B lymphocytes counting, respectively. All procedures were accomplished at Veterinary School, Federal University of Goiás, Brazil. The data were submitted to descriptive statistics and then to Kruskall Wallis and Mann-Whitney tests. The results showed decreased levels of leukocytes, lymphocytes, T lymphocytes and B lymphocytes along the age advance. Absolute values of leukocytes, lymphocytes and T lymphocytes were higher in males than females. None of the evaluated parameters were affected by differences of the management carried out at two farms.Item ANESTESIA COM CETAMINA S(+) ASSOCIADA À ATROPINA E XILAZINA EM CÃES: AVALIAÇÃO CARDÍACA E BIOQUÍMICA SÉRICA(Universidade Federal de Goiás, 2008-03-05) FRANCO, Leandro Guimarães; FIORAVANTI, Maria Clorinda Soares; http://lattes.cnpq.br/8772502020076257; SILVA, Luiz Antônio Franco da; http://lattes.cnpq.br/0446055905975647Changes in physiological parameters of electrocardiography, echocardiography and biochemistry markers were evaluated in dogs anesthetized with different associations of atropine, xylazine and S-ketamine. Twenty three healthy female dogs randomly distributed in four groups named as GI-6, GII-6, GIII-6 and GIV-5 were treated respectively with atropine and S-ketamine (0,04mg/kg; 10 mg/kg); S-ketamine (10 mg/kg); atropine, xylazine and S-ketamine (0,04mg/kg; 1,1 mg/kg; 10 mg/kg) and xylazine and S-ketamine (1,1 mg/kg; 10 mg/kg). Ten minutes after induction, 5mg/kg of S-ketamine was administered to animals from all groups. Measurements of electrocardiogram, echocardiogram and serum activity of aspartate aminotransferase (AST), creatine kinase (CK) and creatine kinase MB isoenzyme (CK-MB) were evaluated for 36 hours. Concerned to electrocardiography, the proposed anesthetic protocols showed significant changes especially in atrioventricular conduction period (PR interval) and systole period (QT interval), mostly in GIV. In relation to echocardiography, the main alterations happened in following variables, final stroke volume, ejection fraction and cardiac output, predominantly in GIV. In serum biochemistry analysis, it was observed alterations in CK and CK-MB values in all groups, which maintained changed for a longer time in GIII and GIV. There were used 384 japanese quails, assigned according to the experimental design (ramdomized blocks), considering the initial live weight of the birds as blocks in the beginning of the experiment. The treatments were three genetic groups with eight replicates, in a total of 48 experimental units with eight birds in each experimental unit (two males and six female). The birds were fed rations according to recommendations of NRC (1994), having soybean meal and ground corn as the main components. The variables of performance were studied every 14 days and three incubation trials were done with 15, 24 and 33 weeks of age. For the statistical analysis the proc GLM of SAS software was used and the means were compared by the t Student test. In each incubation study the completely ramdomized blocks experimental design was used, considering the incubating machines as blocks, with 70 replicates per treatment and one egg as the experimental unit. The parameters evaluated were the egg weight, hatchability, chick weight at hatching, weight loss and weight after six hours of fasting. The results were submitted to variance analysis and the means compared by the t Student test (5% of probability). The genetic groups presented differences among each other for the live body weight in the beginning of production, age at 50% of of productiontwere different to body weight at the beggining and age at 50% of production, where the commercial parent line obtained the best performance. The parameters peak persistence, eggs production, feed intake, viability and beak behavior did not present differences among the genetic groups. The live body weight influenced the egg weight. In the incubation trials the commercial parent line presented more weight of the incubating egg and more weight of the chicks at hatching. It was concluded in the present study that there are statistical differences on the performance and incubating parameters among genetic groups of japanese quails.Item Componentes sanguíneos de bovinos (Bos taurus) da raça Pantaneira em diferentes faixas etárias, criados extensivamente(Universidade Federal de Goiás, 2008-04-11) BORGES, Alinne Cardoso; FIORAVANTI, Maria Clorinda Soares; http://lattes.cnpq.br/8772502020076257The Pantaneiro cattle reveal docility, resistance to illnesses and parasites, and an incredible ability to adapt to Pantanal's edophoclimatic conditions. The cattle may be used without considerable investment to explore low-quality natural pastures in which other breeds might endure low productivity or even survival difficulties. Thus the determination of the normal blood constituents of Pantaneiro bovines was set in order to reveal further information regarding the physiological aspects of these animals. Therefore 293 blood samples were collected from two rural properties, one from Mato Grosso and the other from Mato Grosso do Sul. These individuals were classified in five groups according to their age group, as follows: G1, G2, G3, G4, and G5. The parameters under investigation were the erythrogram and the leukogram. Initially the data was submitted to descriptive statistics which included the determination of averages, standard deviation, and coefficients of variation. Subsequently data was submitted to the Kruskall Wallis test in order to establish the age relationship and to the Mann-Whitney test in order to establish the sex relationship. Age advancement revealed an increase in MCV, MCH, and in the number of eosinophils. Age advancement showed a reduction in the number of red blood cells, hemoglobin, MCHC, segmented neutrophils, and band neutrophils. The values of globular volume and monocytes revealed a rising behavior up to 35 months of age and subsequently suffered a reduction as age advanced. The values of lymphocytes and total leukocytes presented a rising behavior up to 11 months of age and a subsequent decline as age increased. In the first experimental phase, 7224 Dekalb eggs with 32 and 57 weeks old, were incubated with three, five and seven days of storage. The treatments were defined according to the breeder hens ages and their period of storage of eggs, summing up to six treatments with fourteen repetitions of treatment each. The statistical outline was random and the tray height was a covariable. The efficiency of the incubation, the eggs loss of weight during storage, the quality of incubated eggs and the absolute weight of the yolk sac in relation to the weight of the chicken was evaluated. On the second experimental phase, 600 chickens from the six treatments of the first experimental phase were used. The poultry were bred up to the 28th day of life in common batteries. The statistical outline was totally random, and the height of the batteries was a covariable, with five repetitions of twelve birds in each treatment. The starter performance of chicks (up to 28 days of life) was evaluated. The results were submitted to variance analysis and to verify the importance of differences between the average of treatments, the Tukey test was used, with a probability of 5%. The breeder hens of 57 weeks produce heavier eggs with a larger quantity of albumen and yolk. The old breeder hens eggs had a lower quality of albumen when stored for seven days. The loss of weight of the eggs during storage and removal was higher according to the aging of the breeder hen. Eggs produced by the young breeder hens had a higher hatching rate; however, no matter the age of the breeder hen, there was a decrease of the hatching rate of fertile eggs whenever the storage period was increased. The breeder hens with 57 weeks produced heavier chicks and lower chick/egg weight coefficient. The higher ratio yolk sac/chick weight was obtained by the eggs produced by the old breeder hens. There was a lower weight loss of chicks from young breeder hens during the removal of incubatory trays to the grange. The trial was done in Goiânia-GO and lasted 95 days. Five non castrated male lambs from Santa Inês breed were utilized, with an average age of eight months and live weight of 30,2kg ± 4,8kg. The experimental design was a 5x5 Latin Square, the diets were isoproteic (13,5% CP) and isoenergetic (63% TDN) and the forage:concentrate ratio was 60:40. The parameters evaluated were the dry matter intake (DMI) and relative voluntary intake (RVI), apparent digestibility of nutrients, nutritive value index, physically effective fiber, rumen pH and rumen kinetics. The treatments were the following: 100% PSS:0% SCS (100P); 75% PSS:25% SCS (75P); 50% PSS:50% SCS (50P); 25% PSS:75% SCS (25P) and 0% PSS:100% SCS (0P). There were no differences (P>0,05) among treatments for the DMI (597,27 g/day) and RVI (58,41 g of MSU/day). The dry matter digestibility (DMD) and neutral detergent fiber digestibility (NDFD) did not differ (P>0,05) among diets. The crude protein digestibility (CPD) increased when SCS was linearly added, whose values were 68,09%; ,57%; 70,68%; 73,06% and 78,05%, respectively for the treatments 100P, 75P, 50P, 25P and 0P, which can be explained by the addition of 0,5% urea when the sugar cane was ensiled and characterizing the presence of non protein nitrogen (highly soluble in the rumen). It was not verified differences for the nutritive value index (NVI) among treatments, where the mean was 37,74%. Probably due to the DMI and DMD did not present differences (P>0,05) among treatments. Regarding the physically effective fiber (peNDF), there was not effect among treatments and the mean value was 25,92%. There was reduction in the ruminal digesta dry matter disappearance (%/h) with increasing addition of SCS. The values for this parameter were 8,08%/h; 8,31%/h; 5,85%/h; 6,49%/h and 7,03%/h respectively for the treatments 100P, 75P, 50P, 25P and 0P. Since there was no effect in the DMI among treatments and the ruminal digesta dry matter disappearance in the treatments having PSS was higher, it can be concluded that the dry matter digestion rate of the PSS was also higher. The rumen pH increased linearly with addition of SCS, where the values for the treatments 100P, 75P, 50P, 25P and 0P were respectively 6,16; 6,16; 6,33; 6,42 and 6,63. This study aimed to determine the prevalence of antibodies against Leptospira spp., determine their regional distribution and evaluate the main risk factors for the disease in cattle in the State of Goias. The study was conducted using the 4571 samples taken from 715 properties of 213 of the 246 municipalities in the state of Goiás The samples were analysed by the microagglutination test. To determine the correlation between the prevalence and associated factors, the test of Logistic Regression was used through the software SPSS. The percentage of positive samples for at least one of the16 serovars tested was 62.2%, with major prevalence of co-agglutination (40.24%), followed by serovars wolffi (14.53%), hardjo (12.70 %), grippotyphosa (10.55%) and shermani (6.55%). The prevalence of antibodies against Leptospira spp. proved to be associated to the following factors: stratum of production, with a greater prevalence in stractum one, predominantly for meat production; practice of artificial insemination; cattle breed; presence of sheep and goats; presence of capybaras; purchase of cattle in breeding exhibitions and from other properties; rent of pastures in any time of year; presence of maternity paddock and occurrence of abortions. On the other hand, it was not found correlation with the following factors: type of exploitation and creation charged in the property; kind of milking; presence of horses, pigs, dogs, cats, deer and other wild animals; presence of other animals; purchase of animal at fairs and/or auctions; selling of cattle in exhibitions, in fairs and/or auctions, from traders or from other properties; presence of pastures in common with other properties, veterinary care and vaccination against brucellosis, IBR and BVD. Among the etiologic agents identified, there is a predominance of bacteria species. However, several other groups of microorganisms may be present, one of those is a unicellular alga of the Prototheca genus, the Prototheca zopfii, an environmental agent of bovine mastitis. The infected cows usually have granulomatous changes in mammary tissue. Moreover microorganisms do not respond to routine therapy, resulting in the elimination of the infected animals. The main purposes of this study was to determine the occurrence of Prototheca zopfii on bulk tank milk in the state of Goiás and identify the limit of detection for Prototheca zopfii by two different approaches. We also wanted to know the samples pre-incubated with small concentration of Protoheca zopfii permits us to obtain positive growth in microbiological culture. We analyzed 473 samples from farms with high somatic cell count patterns but we did not detect Protoheca zopfii in any of those samples. The limit of sensitivity of the Prototheca zopfii was analyzed, in the approach of isolation the limit of detection was between 130 to 3,400 UFC/mL. In the approach of tally the limit was between 25 to 300 UFC/mL. It wasn t observed significant difference (p>0,05) between the approaches analyzed. However, significant statistical difference (p<0,05) occurred between the five types analyzed. Nile tilapia juveniles (Oreochromis niloticus), Supreme strain, with starter weight of 31.86 ± 1.75 g were randomly allotted in 32 tanks of 100 L of capacity each one in raceway system. The full water exchange of the tanks occurred in 30 min at the beginning. The fish were fed with extruded diet containing 36% of CP in pellets of 2-4 mm in diameter. By the average weight of 100 g, the fish were fed with extruded diet with 32% of CP in pellets of 4-6 mm in diameter until the end of the experiment. This assessment was performed to evaluate feed frequencies (2, 3, 4 and 5 times/day) and ratio (ad libitum and based on the percentage of the biomass weight - %PVBIO) on the productive performance, chemical composition, biometric indices, carcass and fillet yield and costs and profitability partial of Nile tilapia during the weight gain in the raceway system. The experimental design was composed by a completely randomized design in a factorial (4 x 2), with four repetitions, totalizing 32 experimental units. The performances body weight (BW), weight gain (WG), final biomass (FB), feed intake (FI), index of food intake (IF), apparent feed conversion ratio (AFCR), feed efficiency (FE), survival (S%), protein efficiency ratio (PER), specific growth ratio (SGR) and uniformity of the lot (U%) -; fillet weight (FW), carcass yield (CY), fillet yield(FY), biometric indices, and chemical composition fillet (total dry matter (TDM), moisture (MO), crude protein (CP), crude lipid (CL), ash) were the main evaluated parameters. The performance data were submitted to the analysis of variance and Duncan tests (5%), and the others to Tukey test (5%). Despite the quantity of food provided in the treatments, the main variables of water quality remained within the limit recommended for fish breeding. Better AFCR, FE and PER were reached for fish fed three times a day and greater PVBIO% for those ones fed twice daily ad libitum (P <0.05). The values of FBW, FL, FWG, FB, and FI were higher for fish fed four times a day (P <0.05), with the AFCR and PER values similar to those in fish fed three times a day. S%, SGR, CF and U% suffered no influence by the feeding frequency (P> 0.05). Only in IF differences on how to supply the ration (P <0.05) were observed. Fish fed four, five and three times a day had better average of FY, differing from those fed twice a day (P<0.05). The feeding frequency of less than five times had greater CY and VFI, but it did not differ from the other regimens. Difference in form of supply was observed only for RF, and the best average was reached in fish fed ad libitum. Regarding the chemical composition fillet, no differences in ashes were observed (P>0.05). The highest level of CL was found in three times of feeding frequency, which was statistical relevant (P<0.05) in comparison to the others. Interaction was observed for the variables TDM, MO and CP. The cost incidence (CI) was lower in three times a day of feeding frequency with feed supply based on %BW. At the same time, the quality becomes the most important step on food processing chain, since it s essential to guarantee life quality. Then, we purposed this study to verify the effect of private technical assistance on milk quality. The study was carried out during the dry and rain periods of the year, on 40 dairy farms located at the counties of Bela Vista de Goiás, São Miguel do Passa Quatro, Piracanjuba and Cristianópolis, in Goiás State, Brazil. The farms were randomly selected from herds with high results of somatic cell count (SCC) and total bacterial count (TBC). The effect of technical assistance on milk quality was evaluated using the results of milk analyses before and after visits of technicians on the farms. Milk samples were taken from bulk milk tanks and analyzed to somatic cell count (SCC) and total bacterial count (TBC) in electronic devices. We observed a significant decrease on the SCC and TBC after farmers had received technical assistance on milking procedures and mastitis control. The rain period didn t affect the observed results. As the SCC decreased, we observed an increase on SNF values of milk. In Brazil there aren t inform data of possible outbreaks caused by E. coli O157:H7, but this pathogen has been frequently isolated from cattle s feces, and it was recently isolated from bovine carcass at two slaughterhouses in Goiás. The present study has the objective to characterize by ERIC-PCR E. coli and E. coli O157:H7 detected at bovine carcass surfaces and check the power of this methodology to identify different isolates of E. coli and E. coli O157:H7. ERIC-PCR was used to characterize 111 samples, and it was obtained 32 fingerprints separated in 90 isolates of E. coli and eight fingerprints separated in 16 isolates of E. coli O157:H7. From the total of 111 samples, two isolates of E. coli and five of E. coli O157:H7 were non-tipables. The fingerprints varies from one to 18 bands. The discrimination between the samples was high, showing the big power of ERIC-PCR to discriminate isolates from one specie. The experiments were conducted in the poultry experimental facilities of the Veterinary College of the Federal University of Goiás (EV/UFG), located in Goiânia, Brazil. A total of four experiments were carried out to evaluate performance, digestibility and intestinal biometry of broilers fed diets containing viscera and bone meal (FVO) with different moisture levels in processing and feather and blood meal (FPS) with different pressing levels in processing in pre-starter (one to seven days of age) and starter (eight to 21 days of age). In the first experiment, 9% inclusion of FVO with different moisture levels at processing (4, 5 and 6%) in pre-starter rations. In the second experiment, the same moisture levels of FVO were analyzed for starter rations. In the third experiment, 9% inclusion of FPS with different hydrolysis pressing at processing (2,0, 2,5 e 3,0 kg/cm2) in pre-starter ration was tested. In the experiment 4, the same hydrolysis pressing levels were tested for FPS but now at starter rations. The experiment was conducted in a randomized block design, and the criteria to the blocks determination was the battery-cage, with four treatments and five replications of 12 birds each, in a total of 20 experimental units. The statistical analysis was done using UFV/SAEG (2000) and means compared as Tukey test (%5). Variables tested were: live weight, weight gain, feed intake, feed-to-gain ratio, dry matter, nitrogen and ether extract digestibility, dry matter, nitrogen and ether extract retention and biometric indexes of digestive organs. The results indicated that 9% of FVO or FPS didn t affect the performance results. To prestarter phases, the best results were obtained with diets processed with FVO with 5% moisture and FPS with 2,5 kg/cm2 of hydrolysis processing. For starter rations, FVO or FPS didn t affect broiler performance. The poultry by-products processed for animal feeding can be used without performance lost but is important to know the nutritional processing of these ingredients.
