Resumo
The objective of the study is to characterize the semen of tambaqui Colossoma
macropomum during the breeding season and to evaluate the efficiency of four
cryoprotectants added to the extender on sperm cryopreservation of tambaqui
and to define the best protocol for thawing. Three semen samples were collected
at regular intervals from ten males (pool). In the first stage of the experiment the
characterization of semen in natura was performed and it was subjected to the
routine analysis in optical microscopy: motility, vigor, life time and counting the
number of spermatozoa. In the second stage of the experiment cryopreservation
of semen was performed. The cryoprotectants glycerol, dimethylsulfoxide (DMSO),
dimethylformamide (DMF) and methanol were tested at a ratio of 10.0%. For the
assessment of viability and comparison of treatments testing of post-thaw samples
in vivo was performed in three different protocols for each cryoprotectant, in
thawing time 15, 30 and 45 seconds, 30°C temperature, totalizing 12 treatments
with five replicates each. Quantitative variables were subjected to analysis of
variance (ANOVA) and Tukey test while qualitative variables were subjected to the
Freedman test with 0.05 significance. No significant differences (p> 0.05) were
observed for most of the evaluated parameters. The sperm concentration was
higher in the first sampling (14.54 billion sperm /mL) and the lifetime decreased at
the end of the experiment. The cryoprotectant medium containing methanol and
thawing for 30 s was the treatment that showed the best results fertilization
(77.65%/).
