The cattle may be used without considerable investment to explore low-quality natural pastures in which other breeds might endure low productivity or even survival difficulties. Thus the determination of the normal blood constituents of Pantaneiro bovines was set in order to reveal further information regarding the physiological aspects of these animals. Therefore 293 blood samples were collected from two rural properties, one from Mato Grosso and the other from Mato Grosso do Sul. These individuals were classified in five groups according to their age group, as follows: G1, G2, G3, G4, and G5. The parameters under investigation were the erythrogram and the leukogram. Initially the data was submitted to descriptive statistics which included the determination of averages, standard deviation, and coefficients of variation. Subsequently data was submitted to the Kruskall Wallis test in order to establish the age relationship and to the Mann-Whitney test in order to establish the sex relationship. Age advancement revealed an increase in MCV, MCH, and in the number of eosinophils. Age advancement showed a reduction in the number of red blood cells, hemoglobin, MCHC, segmented neutrophils, and band neutrophils. The values of globular volume and monocytes revealed a rising behavior up to 35 months of age and subsequently suffered a reduction as age advanced. The values of lymphocytes and total leukocytes presented a rising behavior up to 11 months of age and a subsequent decline as age increased. The objective of this work was to evaluate in vivo the frequency of expression of active (phosphorylated) form, of p-MEK 1/2 and p-ERK1/2 in all types of ovarian follicles and corpora lutea in different phases of the estrous cycle of cattle. Bovine ovaries were harvested in a local slaughterhouse and the phase of the estrous cycle was determined, considering seven stages, defined as experimental groups: days 1-3 (n = 2), 4-6 (n = 5), 7-9 (n = 5), 10-12 (n = 3), 13-15 (n = 5), 16-18 (n = 5) and 19-21 (n = 5). After processing, the histological sections were subjected to immunohistochemistry using antibodies that specifically recognize the phosphorylated form of MEK 1/2 and ERK 1/2. Histological sections containing corpora lutea were also subjected to immunohistochemistry with antibodies that recognize p-BCL2 (B cell phospho lymphoma 2) and COX-2 (COX-2). The frequency of marked follicles was evaluated for the presence or absence of markup for the two cellular enzymes. The fragments of the corpus luteum were evaluated according to the scores of intensity for positive staining of the four antibodies. The frequencies of the markers studied in the follicles were compared using the chi-square. In preantral follicles, both MEK and ERK 1/2 showed high levels between 16 to 18 days. In the case of p-MEK, the greater activation of primordial follicles and primary held from 7 to 9. In secondary follicles, the less marked for p-MEK occurred in group 4 to 6. Levels of p-ERK began to rise from 13 to 15 days and subsequently decreased. Comparing the frequencies of MEK and ERK, there was statistic difference (p <0.0001) in antral follicles compared to other types of follicles. It was concluded that the intensity of activation of MAPKs MEK / ERK varies with the phases of the estrous cycle and follicular class studied. Since the corpora lutea were evaluated by analysis of variance after transforming the data, it was observed that the proteins studied showed variation in expression profile, in relation to the development, survival and death of the corpus luteum. Since COX-2, a precursor of prostaglandin, is capable of activating the pathways of MEK / ERK 1/2, it reduces the frequency of expression of cell survival factor BCL-2, leading to the corpus luteum at luteolysis. In the last decade, there was a significant advance in research related to the use of tramadol in horses. There is evidence that the drug has significant analgesic effect in horses through mechanisms unclear. The aim of this study was to evaluate clinical and cardiorrespiratory effects and mechanical nociceptive threshold of tramadol, administered intravenously in horses. Moreover, we assessed also the effects of preemptive administration of tramadol on the clinical effects and post-incisional mechanical hyperalgesia. In the first study, seven adult horses were used in a three-way crossover study design in which tramadol was intravenous administered at 1mg kg-1, 2mg kg-1 e 3mg kg-1. Heart rate, respiratory rate, rectal temperature, systolic arterial pressure, diastolic arterial pressure, mean arterial pressure, intestinal motility, head height and behavioral change were recorded before and during 180 minutes after treatment. Mechanical nociceptive threshold from these points were averaged to obtain mean values at each time with von Frey electronic analgesiometer, applying a force in a hoof. Compared to others treatments, the horses that received 3mg kg-1 showed significant increases in respiratory rate and more pronounced behavioral changes. In horses treated with 1mg kg-1, there was a significant decrease in respiratory rate and more pronounced behavioral changes. Motility scores showed transitory decreased in horses treated with 2 mg kg-1. The tramadol did not caused changes in mechanical nociceptive threshold regardless of treatment. In conclusion, the tramadol at doses evaluated produced minimal transient side effects or clinical changes. However has no antinociceptive effect in a model of mechanical stimulus. In a second study, ten horses were divided in two groups. The horses were treated IV with a single dose of 2 mg kg-1 of tramadol (TG) or with a similar volume of sterile water (SG). Heart rate, respiratory rate, rectal temperature, systolic arterial pressure, diastolic arterial pressure, mean arterial pressure, intestinal motility, and behavioral change were recorded before and during 48 hours after treatment. Mechanical nociceptive threshold was measured by von Frey electronic analgesiometer in post-incisional model. To study, a skin incision was made over the thigh region. Compare to saline, tramadol caused transitory increased in mean and diastolic arterial pressure in first hour after treatment. The tramadol was able to decrease a remained time of primary hyperalgesia, but not prevented its appearance in compared to SG. Ahead of the displayed one, this experiment was developed in the Veterinarian Hospital of the Federal University of Goiás, with 26 adult female dogs, castrated and without defined brand, they had been fed with commercial diet ad libitum, during 36 weeks, and had biweekly accompaniment of the nutritional state by means of clinical parameters (body condition score - BCS), morphometrics (abdominal perimeter, thoracic perimeter, thigh perimeter) and for the equations for determination of the percentage of corporal fat and index of corporal mass (IMC1=weightkg/height of withersm; IMC2=weightkg/hight of withers2 m; IMC3=weightkg/spine lengthm; IMC4=weightkg/spine length2 m). The metabolic accompaniment was carried through, monthly, by means of evaluation of the serum biochemistry (glucose, urea, creatinin, albumin, triglycerides, total cholesterol and HDL). The objective of the work was to verify the associations between the morphometrics variables and the serum biochemistries, by means of the multivariate analysis of main components. In the results, bigger association between corporal weight was perceived, thoracic perimeter and of thigh. Moreover, associations between the IMC3 and IMC4 were verified, respectively, by abdominal perimeter and BCS. The percentage of corporal fat was antagonistic to the perimeter of the thigh and corporeal weight and was associated to BCS and IMC4. Related to the serum biochemistry, associations between glucose and creatinin had been observed; and between the total cholesterol and HDL. Antagonism of the albumin with triglycerides, total cholesterol and HDL was verified. Moreover, the percentage of weight gain and the urea texts had been inversely proportional to the concentrations of glucose and creatinin. The present study allowed to conclude that the morphometric, especially the measure of thoracic perimeter and the classification of body condition score constitute efficient methods of monitorament of the corporal conformation, and also, to conclude that, dogs submitted to the weight gain for a short period of time, had been capable to develop compensatory mechanisms of keeping the homeostasis of metabolism, keeping the biochemical concentrations inside of the values of reference.Item Produção lacrimal e densidade de células caliciformes conjuntivais em cães da raça SHIH-TZU(Universidade Federal de Goiás, 2008-02-28) LIMA, Aline Maria Vasconcelos; ARAÚJO, Eugênio Gonçalves de; http://lattes.cnpq.br/3919777570059928; BRITO, Luiz Augusto Batista; http://lattes.cnpq.br/3212353728321020; CHAVES, Nilo Sérgio Troncoso; http://lattes.cnpq.br/9218281815823682Considering the relevance of tear production and tear quality to the conservation of the corneal health, the present study aimed to evaluate tear production and conjunctival goblet cells density in Shih-tzu dogs. Thirty five grownup Shih-tzu dogs, including 33 females and 2 males, were subjected to Schirmer tear test 1 (STT-1) and Schirmer tear test 2 (STT-2). Among these dogs, fourteen females and one male were underwent to conjunctival biopsy for histologic evaluation of goblet cell density (GCD) and goblet cell index (GCI). The STT-1 mean values found were 19,66 ± 7,3 mm/min for left eye (LE) and 21,97 ± 5,7 mm/min for right eye (RE). The STT-2 mean values were 10,71 ± 6,1 mm/min and 9,14 ± 4,78 mm/min for LE and RE, respectively. The GCD mean values found were 13,64 ± 3,44 goblet cells/50 epithelial cells for LE and 13, 64 ± 4,07 goblet cells/50 epithelial cells for RE. The GCIs correspondent were 0,27 ± 0,069 and 0,27 ± 0,081 for LE and RE, respectively. There was significant difference between LE and RE STT-1 values. There was not significant difference between eyes for STT-2, GCD and GCI values. STT-1 and STT-2 values did not influence significantly GCI values. STT-1 and STT-2 values are within of the pattern considered normal, but the GCD and GCI mean values are lower than those values described in literature for healthy dogs of other breeds. The conjunctival specimens histological evaluation was accepted as convenient, which demonstrated squamous metaplasic areas in 83,3% of the specimens. Within 44% of these, squamous metaplasia was predominantly associated with mononuclear inflammatory infiltrate.Item Avaliação clínico-laboratorial,histopatológica hepática e desempenho de ovinos alimentados com feno de braquiária ou cana-de-açúcar(Universidade Federal de Goiás, 2009-03-03) LIMA, Flávia Gontijo de; ARAÚJO, Eugênio Gonçalves de; http://lattes.cnpq.br/3919777570059928; FIORAVANTI, Maria Clorinda Soares; http://lattes.cnpq.br/8772502020076257The species of Brachiaria are important forages from Brazilian tropical regions, mainly the Central-Western region. Some species of Brachiaria have been described as cause of hepatogenous photosensitization in ruminants. Initially, the disease was attributed to the fungus Pithomyces chartarum, but recent studies suggest that the steroidal saponins present in the grasses have toxic principles responsible for the photosensitization. The objective of this study was to evaluate the hepatic function and the performance of lambs fed with B. brizantha hay or sugar cane (Saccharum officinarum L.), by means of clinical examination, laboratory tests, and macro and microscopic analysis of the liver. Twelve Saint Ines lambs were used. The animals were divided into two experimental groups: group hay (six lambs fed with roughage of B. brizantha hay and concentrate) and group sugar cane (six lambs fed with roughage of sugar cane and concentrate). The hay used to feed the lambs did not contain Pithomyces chartarum spores. The clinical examinations occurred at each seven days, the laboratory tests at each 14 days, and the weighings at each 28 days. At the end of 93 days of experiment the lambs were slaughtered, the macroscopic analysis of the the organs was carried out, and the liver fragments were collected for the microscopic analysis. The lambs were clinically healthy, during the whole period, except at the beginning of the experiment, when some animals presented pneumonia. The only biochemistry alterations suggestive of hepatic damage were the increase of the GGT values and the decrease of total cholesterol in both groups. No animal fed with B. brizantha hay presented macroscopic alteration in the liver. The microscopic analysis of the liver revealed preserved hepatocytes and presence of multifocal infiltration of mononuclear inflammatory cells in the hepatic parenchyma and also in the portal triads, characterizing cholangitis in both groups. Degenerations suggestive of hepatic esteatosis were observed in four animals fed with sugar cane. Feeding lambs with B. brizantha hay promoted similar performance than feeding the animals with sugar cane. Regardless of the type of feeding, the lambs presented, as biochemistry alteration of the hepatic function, increase of the serum levels of GGT and decrease of total cholesterol, followed by histological alterations, characteristic of light cholangitis.Item ESTUDO CLÍNICO E RADIOGRÁFICO DAS EXTREMIDADES DISTAIS DOS MEMBROS LOMOTORES DE BOVINOS MACHOS DE CORTE(Universidade Federal de Goiás, 2009-04-07) LIMA, Ingrid Rios; BORGES, Naída Cristina; http://lattes.cnpq.br/9181279951885005Radiographic evaluation has been done lately as important diagnosis recourse for bovine locomotor injuries. However the scarcity of study in this area has made difficult the veterinary s performance. The aim of the study was to use radiographic exams to help in the characterization of the most important bone injuries that compromise bovine digit, metacarpophalangeal and metatarsophalangeal joint as well as relate radiographic findings to lameness sings and the presence of feet injuries. Participate in this study fifty male bovines (Zebu x European), confined, carrying locomotor alterations, with or without digital alteration, from the state of Goiás. All the animals were evaluated about the locomotion score the moment they arrived, and 112 days latter at the end of the confinement. After the slaughter, the four member distal extremities were collected (n=200) and evaluated about clinic and radiographic alterations. Claudication was found out in 86% of the animals. The locomotion scores were distributed as fallowing: 14% had score 1, 20% score 2, 52% score 3 and 14% score 4. The comparison between two moments, arrival and end of the confinement, demonstrated the aggravation of the locomotion score during the period the animals were confined. Only 29,5% from the extremities evaluated presented digital injuries. From these, 40,7% were classified as mixture, 32,2% septic pododermatitis, 11,9% interdigital dermatitis, 8,5% horn erosion and 68% as digital dermatitis. According to the radiographic evaluation 25,5% of the extremities presented some kind of digital alteration. The main injuries were distributed in joint degenerative disease (27,5%), osteitis (19,6%), periostitis (15,7%), septic arthritis in distal interphalangeal joint (11,8%), calcification off deep flexor tendon (7,8), calcification of the extensor tendon (5,9%), fracture in chip, ankylosis of joint and calcification of the proximal insertions of the cruciate ligaments (3,9% each one). The results showed relation between the presence of lameness and digital injuries with the radiographic alterations observed in the digits, however this relation was not found in the radiographic alterations of the fisis from metacarpophalangeal and metatarsophalangeal joints. We concluded that the locomotion score, associated to characterization of the podal injuries and the radiographic examination, helped in the diagnosis of lameness and that there is a relation among lameness, pedal injuries and radiographic alterations. It also showed that the confinement may be a determinating factor to the development of fiseal dysplasia in bovines, however, for better comprehension of the illness more radiographic studies that go along with the animal growth must be carried outItem AÇÃO DA CLORPROMAZINA EM RINS DE CÃES SUBMETIDOS À ISQUEMIA E REPERFUSÃO(Universidade Federal de Goiás, 2009-09-19) MENEZES, Liliana Borges de; ARAÚJO, Eugênio Gonçalves de; http://lattes.cnpq.br/3919777570059928Renal ischemia is present in different situations such as vascular or kidney surgery and also in renal transplantation. The aim of this study was to evaluate the renal function in dogs submitted to ischemia and reperfusion after chlorpromazine application. Twelve adult mongrel dogs were distributed into two groups with six animals: group A - ischemia and reperfusion without previous administration of chlorpromazine; group B - ischemia and reperfusion treated previously with chlorpromazine. Blood and urine samples were harvested in four different times: before ischemia, at the beginning of reperfusion, 120 minutes after of reperfusion and every week until postsurgical day 28 in order to check the possible ischemia/reperfusion late effect. Renal function was evaluated by clinical examination, serum urea and creatinine levels and urinary GGT activity. PU/CU and urinary activity of GGT were more sensitive for detecting acute tubular injury then routine urine examination, because these variables showed earlier alterations. It was not possible to verify the chlorpromazine protective action through urinalysis, creatinine and urea serum levels, excretion of GGT urinary and urine protein:creatinine ratio.Item Enfermidades Cutâneas Digitais Bovina: Aspectos Genéticos E Terapêuticos(Universidade Federal de Goiás, 2011-03-18) MOURA, Maria Ivete de; FIORAVANTI, Maria Clorinda Soares; http://lattes.cnpq.br/8772502020076257; SILVA, Luiz Antônio Franco da; http://lattes.cnpq.br/0446055905975647This study has aimed do evaluate a risk factor for bovine digital dermatitis and the efficacy of 5% aqueous barbatimão extract solution as ancillary in interdigital skin incisional wound repair. One hundred bovine females, 50 Holstein and 50 Girolando, from two different farms were used for the risk factor study. Experimental units were divided into four groups: healthy Holstein cattle group (GHS) and digital dermatitis Holstein cattle group (GHL), healthy Girolando cattle group (GGS) and digital dermatitis Girolando cattle group (GGL). Ten milliliters of blood were collected from all animals and stored in anticoagulant added tubes. Samples were refrigerated and submitted to hemolytic tests for eight bovine blood systems: A, B, C, F, J, L, S and Z. The frequency of blood factors was determined and compared, in each system and each group. Data were analyzed through non parametric tests: Chi Square and Fisher s exact. The results indicated that the blood factors may contribute to the occurrence of digital dermatitis lesions, but further studies are needed for a more accurate conclusion. Thirty confined, 10 months old calves were selected in order to evaluate the efficacy of 5% barbatimão solution. A biopsy punch was introduced in each interdigital space, up to the dermis, to produce a lesion measuring one centimeter in diameter. The lesions were numbered 120 in total and named experimental units, which were divided into two groups of 60 units each: GB treated with 5% aqueous barbatimão bark extract solution and GC treated with water. Tissue repair was evaluated through macroscopic and histological findings in days three, seven, 14 and 21 of treatment. Macroscopic findings were analyzed descriptively, while histological findings were submitted to Mann Whitney test, with p<0.05. For lesions treated, GB group showed less intense polymorphonuclear cells infiltration on the first 14 days of treatment. A marked collagen deposition was also observed at day 14 while collagen alignment was more expressive on day 21. More specific studies may be necessary in order to determine if blood factors can contribute to the occurrence of digital dermatitis. Barbatiman 5% footbath solution for confined cattle helps qualitatively in modulation of interdigital cutaneous incisional wound repair, but does not avoid the extension of inflammatory phase of healing.Item ATIVIDADES ANGIOGÊNICA, ANTIINFLAMATÓRIA, CICATRIZANTE E ANTIBACTERIANA DO EXTRATO ETANÓLICO E FRAÇÕES DAS FLORES DA Calendula offIcinalis L. Displays orange or yellow flowers, which are attributed medicinal properties, with emphasis on healing activity. In this study, the ethanolic extract of flowers of C. officinalis grown in Brazil was prepared by soaking the cold and fractionated to obtain fractions of dichloromethane and hexane. The angiogenesis activity of the extract and fractions was evaluated using the model of membrane corioalantóide in embryonated chicken eggs. But the healing effect of the extract was evaluated by the model of skin wounds in rats, with assessment of macroscopic and microscopic characteristics. In this model, the ethanol extract of angiogenesis activity in the dermis of rats was also evaluated by immunohistochemistry, and measured the intensity of expression of vascular endothelial growth factor (VEGF). The in vitro antibacterial activity of extract and fractions was evaluated by determining the minimum inhibitory concentration (MIC) using the dilution plate. The ethanol extract and the fractions of calendula showed marked angiogenesis activity, demonstrated in both experimental models used. This activity was not related to the observed increase in intensity of expression of VEGF, which may be related to other pro-angiogenic factors. In the model of skin wounds in rats the ethanolic extract showed anti-inflammatory activity, demonstrated by the decrease in serous exudation of hyperemia, the deposition of fibrin and epidermal hyperplasia, besides the presence of more thin crusts and soaked in the control group. There was an increase in the amount of collagen in granulation tissue and antibacterial activity was observed, mainly on gram-positive bacteria. Still, the C. officinalis served as positive about the process of vascular neoformation and activity in healing skin wounds in rats, and also presented antibacterial activity in vitro. The exams were carried out in a i-CAT tomograph, using for the images acquisition six centimeters height, 40 seconds time, 0,2 voxel, 120 kilovolts (kV) and 46,72 milliampéres per second (mAs). The ideal positioning of the animal for the exam was also determined in this study and it proved to be fundamental for performing the exam, which required a simple and safe anesthetic protocol due to the relatively short time necessary in obtaining the images. The immunogenicity of recombinant MPT-51 (rMPT-51), Ag-85 and M. bovis-BCG were characterized in an immunosorbent assay, where 208 serum samples from positive intradermal tuberculin test (ITT) animals and 54 serum samples from ITT negative animals where analyzed. M. bovis-BCG and Ag-85 were strongly recognized by antibodies from naturally infected cattle. Additionally, the clinical status of the animals were correlated with the ITT positivity, with the specific production of IL-4 by TCD4 and TCD8 positive lymphocytes, and with nitric oxide (NO) production by macrophages from naturally tuberculosis infected bovine peripheral blood. ITT positive animals showed TCD4+IL4+ cells specific to M. bovis-BCG extract. High background levels of TCD8+IL-4+ lymphocytes were observed in ITT positive animals independent of the stimuli. When cell cultures where stimulated with M. bovis-BCG protein extract, there was no observed difference in NO production between the groups. Naturally tuberculosis infected bovine presented TCD4+IL4+ cells specific for M. bovis- BCG and a preserved NO production. Finnally, the immunogenicity of rMPT-51 use as a proteic sub-unit vaccine was evaluated in BALB/c mice, with two different adjuvants, incomplete Freund and CpG DNA. For this, mice were immunized and challenged with M. tuberculosis. These animals are constant victims of running over in highways and still they suffer with intense infestation for parasites, what it can result in intense anemia, being necessary the accomplishment of blood transfusion. In the present study blood samples were used of eight jaguarundies (Puma yagouaroundi), eight ocelots (Leopardus pardalis), seven pampas cats (Leopardus colocolo), seven domestic cats (Felis catus) of the Persian race and eight domestic cats (Felis catus) without definite race (SRD), whose aim was: a) to carry through blood typing and tests of compatibility between corresponding blood types in the different species; b) to carry through count blood cells (CBC), comparing the technique automatized by means of device BC - 2800 vet (software for domestic cats), with the manual technique for the wild species; and c) to carry through the profile biochemist and electrophoretic of serum proteins. In the typing, the occurrence of the blood type A was of 100% between ocelots, pampas cats and domestic cats Persian and of 85,72% between domestic cats SRD. 100% of the jaguarundies were type B and 14,28% of domestic cats SRD. To the tests of blood compatibility, 87,5% (n=4) of the ocelots were incompatible with domestic cats and 12,5% (n=1) were compatible; 100% (n= 6) of the pampas cats were compatible with domestic cats and 100% (n= 4) of the jaguarundies were incompatible with domestic cat of type B. It is concluded that in accordance with tests of blood compatibility the transfusion between domestic and pampas cats is possible, it is impossible the transfusion between domestic and jaguarundies and that more studies must be carried through for ocelots. In the comparison enters the techniques for the accomplishment of the CBC the automatized technique demonstrated statistical results equal for the majority of the parameters, also inside of the values of reference for the species, except in the counting of leukocytes of jaguarundies, whose resulted gotten for automation were bigger. It had good correlation between the techniques, mainly in the ocelot species. Concludes that device BC 2800 vet with the configuration for domestic cats is a technique fast and trustworthy in the accomplishment of blood cells count for species pampas cat and ocelot. The manual counting of leukocytes must be preferred for jaguarundies. The results gotten for many of the evaluated parameters biochemistries were similar to the described as values of reference for the studied species. However, the values found for ALT and AST were above of the values of reference for jaguarundies and ocelots. The value of glucose was above for the species pampas cat. The urea was above of the values of reference for the three species. The electrophoretic race for the analyzed species was similar to the described one for domestic cats contends albumen, alpha globulins (alpha 1 and 2), beta globulins (beta 1 and 2) and gamma globulins, however, in all the species had individuals that they had not presented beta globulin of the type beta 2. The gotten results can be used as normality parameters for the species pampas cat, ocelots and jaguarundies from Cerrado biome.
