| dc.description.abstract | The design of organic fluorophores with tailored optical properties is critical for advancing technologies in optoelectronics and bioimaging. Among them, 2,1,3-benzothiadiazole (BTD) derivatives stand out due to their strong π-conjugation, high fluorescence efficiency, and ability to form ordered structures. However, rationally predicting their photophysical properties remains challenging due to the complex interplay between substituent effects, solvent polarity, and electronic transitions. Traditional methods relying on quantum chemistry are accurate but computationally expensive, limiting their scalability for high-throughput compound screening. To address this gap, this work proposes a data-driven strategy combining machine learning (ML) and explainable artificial intelligence to accelerate the prediction and interpretation of photophysical properties in BTD derivatives. The primary objective was to develop robust ML models for predicting maximum absorption and emission wavelengths and to derive mechanistic insights via SHapley Additive exPlanations (SHAP). The methodology involved curating a dataset of BTD derivatives, generating molecular descriptors through Morgan fingerprints, and training models using Random Forest, LightGBM, and XGBoost algorithms. Model validation employed 10-fold crossvalidation and y-randomization, with performance assessed via R2, Q2, MAE, and RMSE metrics. The results demonstrated that ML models achieved high predictive accuracy, with the Random Forest model outperforming others for both photophysical endpoints. SHAP analysis identified electron-donating groups, particularly tertiary amines, and solvent polarity as key determinants of optical behavior. The deployed web tool enables real-time property prediction and substructure attribution, thus bridging computational chemistry and practical compound design. These findings underscore the potential of interpretable ML to guide the rational design of organic materials, reducing reliance on expensive calculations and fostering accessibility for chemists and material scientists. | eng |
| dc.description.resumo | O desenvolvimento de fluoróforos orgânicos com propriedades ópticas ajustáveis é essencial para o avanço de tecnologias em optoeletrônica e bioimagem. Entre eles, os derivados da 2,1,3-benzotiadiazol (BTD) se destacam por sua forte conjugação π, alta eficiência de fluorescência e capacidade de formar estruturas ordenadas. No entanto, prever racionalmente suas propriedades fotofísicas ainda é um desafio, devido à interação complexa entre efeitos de substituintes, polaridade do solvente e transições eletrônicas. Métodos tradicionais, baseados em química quântica, são precisos, mas computacionalmente dispendiosos, dificultando sua aplicação em triagens de compostos em larga escala. Para preencher essa lacuna, este trabalho propõe uma estratégia baseada em dados que combina machine learning (ML) e inteligência artificial explicativa para acelerar a previsão e a interpretação das propriedades fotofísicas de derivados de BTD. O principal objetivo foi desenvolver modelos preditivos robustos para os comprimentos de onda de absorção e emissão máximos, além de obter conhecimento mecanísticos por meio da técnica SHapley Additive exPlanations (SHAP). A metodologia envolveu a construção de um conjunto de dados com derivados de BTD, geração de descritores moleculares por meio de morgan fingerprints (MF) e o treinamento de modelos usando os algoritmos Random Forest, LightGBM e XGBoost. A validação foi realizada com validação cruzada (10-fold) e y-randomization, avaliando o desempenho com métricas como R2, Q2, MAE e RMSE. Os resultados mostraram que os modelos de ML apresentaram alta acurácia preditiva, com destaque para o Random Forest, que superou os demais em ambas as propriedades estudadas. A análise SHAP identificou grupos doadores de elétrons, especialmente aminas terciárias, e a polaridade do solvente como determinantes chave no comportamento óptico. A ferramenta web desenvolvida permite a previsão em tempo real e a visualização das contribuições estruturais, promovendo a integração entre química computacional e design racional de compostos.
