dc.description.abstract | Plant biodiversity is considered the main source of secondary metabolites useful for chemical synthesis. Among the different classes of plant metabolites with therapeutic properties are chalcones. This group represents an important class of organic compounds with numerous biological activities, such as anti-inflammatory, anti-HIV, antibacterial, antifungal and anticancer. This study aimed to investigate the angiogenic, antiangiogenic, mutagenic and antimutagenic potential of bischalcone synthetic αβ7E, 4E-1- (4-chlorophenyl) -5- (2,6,6-trimethylcyclohexen-1-yl) penta-1,4-dien -3-one (CAB7β), besides its optical properties. The angiogenesis test with the chorioallantoic membrane (CAM) of embryonated chicken eggs (Gallus domesticus) with the S180 sarcoma tumor line, the Ames and micronucleus mutagenicity tests in mouse bone marrow, and the techniques spectroscopic two-photon absorption (2PA), visible ultraviolet (UV-vis) and Z-Scan, respectively. In the angiogenic test, CAB7β promoted a significant reduction (p <0.05) in the total vessels, junctions, length and caliber of the blood vessels stimulated by the S180 cells, besides reducing the presence of the inflammatory elements in the vessels of the CAMs. Another observation was that MCAs treated with CAB7β only showed significant reductions (p <0.05) in the angiogenic process compared to the negative control group used in this study. In the Ames test, CAB7β showed a protective effect on DNA against the mutagenic action of 4-nitroquinoline-1-oxide and sodium azide, and also against the action of doxorubicin in co, pre and post-treatment models using the micronucleus test. CAB7β at the doses of 12.5 and 50 μg / plate presented a moderate cytotoxic profile in TA98 strain of S. typhimurium. In the micronucleus test, its higher dose (50mg / kg p.c.) also exhibited cytotoxic activity. However, an anti-cytotoxic pertituent of this compound was observed at all doses tested in TA100 and TA-98 strains of S. typhimurium and during co, pre and post-treatment in the micronucleus test. The experimental and theoretical results of the optical properties of CAB7β demonstrated that the absorption spectrum of this compound has an intense absorption in the region of the UVA, which is associated to a transition of the type π → π * between the HOMO - LUMO orbitals, with a transfer intramolecular charge ratio of the trimethylcyclohexen-1-yl ring to the chlorophenyl ring. Thus, we conclude that the results of this study demonstrated, therefore, the versatility of CAB7β. This molecule may be a promising target in the development of new therapies against cancer, especially against sarcoma, due to the antiangiogenic potential associated with the reduction of inflammatory elements and their significant anti-mutagenic and anti-cytotoxic effects. dc.description.resumo | A biodiversidade vegetal é considerada a principal fonte de metabólitos secundários úteis para a síntese química. Dentre as diferentes classes de metabólitos vegetais com propriedades terapêuticas estão as chalconas. Este grupo representa uma importante classe de compostos orgânicos com inúmeras atividades biológicas, tais como anti-inflamatória, anti-HIV, antibacteriana, antifúngica e anticâncer. Este estudo objetivou investigar o potencial angiogênico, antiangiogênico, mutagênico, antimutagênico da bischalcona αβ7 sintética 1E, 4E-1- (4-clorofenil) -5- (2,6,6-trimetilciclohexen-1-il) penta-1,4-dien-3-ona (CAB7β), além de suas propriedades ópticas. Para tanto, foi utilizado o teste de angiogênese com a membrana corioalantóide (CAM) de ovos embrionados de galinha (Gallus domesticus) com a linhagem tumoral do sarcoma S180, os testes de mutagenicidade de Ames e do micronúcleo em medula óssea de camundongos e as técnicas espectroscópicas de absorção de dois fótons (2PA), ultravioleta visível (UV-vis) e Z-Scan, respectivamente. No teste angiogênico, a CAB7β promoveu uma redução significativa (p < 0,05) do total de vasos, junções, comprimento e calibre dos vasos sanguíneos estimulados pelas células S180, além de reduzir a presença dos elementos inflamatórios nos vasos das CAMs. Outra observação foi que as MCAs tratadas apenas com a CAB7β apresentaram reduções significativas (p < 0.05) no processo angiogênico em comparação ao grupo controle negativo utilizado neste estudo. No teste de Ames a CAB7β apresentou efeito protetor ao DNA diante da ação mutagênica da 4-nitroquinolina-1-óxido e azida sódica, e também contra a ação da doxorrubicina nos modelos de co, pré e pós-tratamento pelo teste do micronúcleo. A CAB7β nas doses de 12,5 e 50 µg/placa, apresentou um moderado perfil citotóxico na cepa TA98 de S. typhimurium. No teste do micronúcleo, sua maior dose (50mg/Kg p.c) também exibiu atividade citotóxica. No entanto, um pertil anticitotóxico deste composto foi observado em todas as doses testadas nas cepas TA100 e TA-98 de S. typhimurium e durante o co, pré e pós-tratamento no teste do micronúcleo. Os resultados experimentais e teóricos das propriedades ópticas da CAB7β demonstraram que o espectro de absorção deste composto possui uma intensa absorção na região do UVA, a qual está associada a uma transição do tipo π→π* entre os orbitais HOMO – LUMO, com uma transferência intramolecular de carga do anel trimetilciclohexen-1-il para o anel clorofenil. Assim, concluímos que os resultados deste estudo demonstraram, portanto, a versatilidade da CAB7β. Esta molécula pode ser um alvo promissor no desenvolvimento de novas terapias contra o câncer, em especial contra o sarcoma, devido ao potencial antiangiogênico associado à redução de elementos inflamatórios e a seus significativos efeitos antimutagênico e anticitotóxicos. Além disso, de apresentar potencialidades para o desenvolvimento de filtros fotoprotetores e na terapia fotodinâmica dada suas propriedades ópticas. | eng |
