dc.description.abstract | The essential oil of leaves of the Eucalyptus microcorys F. Muller collected in Silvânia-GO, possesses 87% of 1,8-cineole. This monoterpene is one of the main composed of the essential oils of Eucalyptus globulus Labill and they are of great interest for the industries, mainly the perfumeries, food and pharmaceutical. The objective of this work was the hydroxylation of 1,8-cineole for free and immobilized bacterias. The hidroxilation of the molecule of 1,8- cineole is important for obtaining new aromas. The essential oil of Eucalyptus microcorys also presented inhibition on the growth of Staphylococcus aureus ATCC 6538 and the hydroxylation of the molecule of 1,8-cineole can increase this activity antibacterial. In this work it was tested the Pseudomonas putida, Azotobacter vinelandii and Pseudomonas fluorescence bacterias. However the Pseudomonas putida went to that presented a significant result and facilitated the biotransformation of the composition 1,8-cineole. The main culture used for biotransformation (MMB) was: 0,5g of glucose, 0,2g of (NH4)2SO4, 0,4g of KH2PO4, 0,6g of Na2HPO4, 0,2g of MgSO4, amount lines of Fe, B, Mn, Zn, Cu and Mo in 100 mL of distilled water. After 60-65 hours to 30oC, pH 6,5 and 180 rpm 100 μL the 1,8-cineole was added in main MMB and was maintained at the same temperature conditions, pH and agitation, for periods of 48-120 hours. The solvent that showed more efficient for extraction of 1,8-cineole was the ethyl acetate. The ideal revelator for the composition 1,8-cineole at thin layer chromatography was a solution of 10 mL of H2SO4 concentrated, 45 mL ethanol, 1g of vanillin and 10 mL of distilled water. The mobile phase used at the chromatography was 1:3 of ethyl/hexano acetate. The results of biotransformation were investigated through the technique of GC/MS. The bacteria Pseudomonas putida got transform the substratum 1,8-cineole in two new composed: exo-2-hydroxycineole and the 1- hydroxy-p-menth-3-one. | pt_BR |
dc.description.resumo | O óleo essencial das folhas dessecadas de Eucalyptus microcorys F. Muller coletadas em Silvânia-GO, possuem 87% do monoterpenóide 1,8-cineol. Este monoterpeno é um dos principais compostos do óleo essencial de Eucalyptus globulus Labill de grande interesse para as indústrias, principalmente as de perfumaria, alimentos e farmacêutica. O objetivo do trabalho foi a hidroxilação do 1,8-cineol por bactérias livres e imobilizadas. A hidroxilação da molécula do 1,8-cineol é importante para obtenção de novos aromas. O óleo essencial de Eucalyptus microcorys também apresentou inibição sobre o crescimento de Staphylococcus aureus ATCC 6538 e a hidroxilação da molécula do 1,8-cineol pode aumentar esta atividade. Neste trabalho utilizou-se as bactérias Pseudomonas putida, Azotobacter vinelandii e Pseudomonas fluorescence. Entretanto a Pseudomonas putida foi a que apresentou melhor resultado na biotransformação do 1,8-cineol. O meio MMB utilizado para biotransformação foi desenvolvido com a seguinte composição: 0,5g de glicose, 0,2g de (NH4)2SO4, 0,4g de KH2PO4 , 0,6g de Na2HPO4, 0,2g de MgSO4 , quantidade traços de Fe, B, Mn, Zn, Cu e Mo em 100 mL de água destilada. Após 60-65 horas a 30oC , pH 6,5 e 180 rpm adicionou-se 100 μL do 1,8-cineol no meio MMB sendo mantido nas mesmas condições de temperatura, pH e agitação, por períodos de 48-120 horas. O solvente que mostrou mais eficiente para extração do 1,8-cineol foi o acetato de etila. O revelador para o composto 1,8-cineol na cromatografia de camada delgada foi a vanilina sulfúrica constituída por uma solução de 10 mL de H2SO4 concentrado, 45 mL de etanol, 1g de vanilina e 10 mL de água destilada. A fase móvel usada na cromatografia foi hexano/acetato de etila (75:25). Os resultados da biotransformação foram investigados através da técnica de CG/EM. A bactéria Pseudomonas putida conseguiu transformar o substrato 1,8-cineol em dois novos compostos: hexo-2-hidroxicineol e o 1-hidroxi-p-ment-3-ona. | pt_BR |
