Resumo
The Alzheimer's Disease (AD) is a progressive neurodegenerative disorder manifested by the
deterioration of memory associated with neurofuncional decline, behavioral disorders and psychological symptoms.
The AD has a heterogeneous etiology with several possible causes, including genetic susceptibility, metabolic
abnormalities, abnormal processing of proteins, and deficits of neurotransmitters. It is known that the AD is a
condition of high frequency among patients older than 65 years of age. The diagnosis is based on clinical
characteristics; however, a definitive diagnosis requires the histopathologic examination of the brain after the
patient’s death. The several studies and reviews on AD either focus on only one class of drugs, a specific
pharmaceutical group, psychological characteristics of the disease or on biochemical characteristics, however, this
review discussed the actions of various drugs and their partial effectiveness, because drugs that have effective
action against such dementia have not yet been discovered. The objective of this paper was also to associate
information about the various drugs used to treat AD with focus on those that act on the cholinergic
neurotransmission metabolism, glutamate exotoxicity and phytotherapy involving them and other metabolic action
mechanisms, as well as to discuss some aspects that may interfere with the search for discoveries of more
effective medicines for the treatment of AD, taking into account possible psychosocial and ethical problems. In
conclusion, drugs formulated for the treatment of the AD of anticholinergic, memantine and phytotherapy classes
do not prevent the development of dementia and have palliative effects. It is possible that the psychosocial and
ethical aspects of researchers involved in the sphere of this dementia impair the discovery of new techniques for
the treatment of the AD.
